Discovery of a novel DNA repair system

This work is published in Science and was performed by Gilbert Richarme and his collaborators from CNRS, CEA, universities Paris-Diderot, Paris-Descartes, Paris-Sud1 and from the Harvard Medical School, and the Lebanese American University .

“Bacterial systems are invaluable for discovering the molecular bases of living organisms. Our new findings use the model bacterium Escherichia coli to understand the function of a human protein called DJ1, which is implicated in some cases of Parkinson’s disease. This protein has three functional homologues in E. coli. Glyoxals are harmful byproducts of sugar metabolism that damage cellular components. The team of Prof. Gilbert Richarme had previously shown that DJ1 and its bacterial homologues are deglycases that repair proteins altered by glyoxals. The new work in which we collaborated shows that DJ1 and its bacterial counterparts repair guanines damaged by glycation, whether they are free or incorporated in DNA. Deficient bacteria with no deglycases have an increased mutation rate. This discovery highlights a system conserved from bacteria to humans that preserves DNA integrity.”
