    Insights on the 4D dynamics of budding yeast mitotic and meiotic chromosomes

    Résumé : Abstract
    The duplication and segregation of chromosomes during mitosis and meiosis involve the dynamic re-organization of their internal structure by conserved architectural proteins, such as structural maintenance of chromosomes complexes (i.e. cohesin and condensin). Although the roles of these factors are actively investigated, a genome-wide overview of chromosome dynamic architecture at both small and large-scales during these two essential processes remain to be reached. I will present recent work on budding yeast : first, the comprehensive 4D analysis of higher-order genome organization during the mitotic cell cycle, and the roles of SMC in the observed structural transitions. Then, a preliminary investigation of homologs dynamics during meiosis prophase using Hi-C, a study which necessitate the redesign and reassembly of a 145kb region. We track in a synchronized meiotic population the establishment of homolog pairing, and the individualization, condensation, and internal restructuration of paired chromosomes.
