      RNase H deficiency reveals the existence of mysterious R-lesions in DNA

      Résumé : RNase H-deficient E. coli (rnhAB mutants, lacking both H1 and H2 enzymes) grow extremely slowly. We show that this slow growth is a combination of regular formation of repairable chromosomal lesions with periodic formation of irreparable ones. The rnhAB mutants of E. coli depend on recombinational repair and fragment their chromosomes, but the double-strand DNA breaks appear to be the consequence of the inability of this mutant to remove ribonucleotides from DNA — in short, to repair the so-called R-lesions. Inhibition of translation and DNA replication is detrimental for the rnhAB mutants, while inhibition of transcription is beneficial for them, suggesting involvement of R-loops in generation of R-lesions. Our genetic and physical investigation shed further light on the possible nature of these mysterious DNA lesions.

      Drug resistance in the malaria parasite Plasmodium falciparum - the case of atovaquone-proguanil

      Résumé : Antimalarial drugs are one of the main tool for the control of malaria, but malaria parasites have acquired resistance to nearly all clinically available drugs since several decades. After a presentation of the main antimalarial chemotherapeutic agents, I will focus on resistance to atovaquone-proguanil that associates with late treatment failures through de novo mitochondrial mutations. I will present the mode of action of this drug, and the mechanism and evolution of drug resistance in patients. Various methods have been employed, including genetics, pharmacology, computational modeling, statistical analysis of patient’s databases, and the yeast model.
